Les objectifs de cette réunion étaient de faire un bilan des activités 2013 du Laboratoire National de Référence (LNR), des réseaux de surveillance (REPAMO) et d'Observation (RESCO et réseaux de centres techniques).

Among the Iridovirus related infections frequently described on many teleosts fish species, Viral Erythrocytic Necrosis (VEN) (Pinto et al., 1989), Epizootic Haematopoietic Necrosis Virus (EHNV) (Langdon et al., 1988) as weil as Lymphocystis Disease Virus (LDV) are not considered to directly cause fatality. Rapid and sensitive detection of ostreid Herpesvirus 1 in oyster samples by real-time PCR (Poster) ArchiMer

Herpes and herpes-like virus infections have been reported in various marine mollusc species associated with high mortality rates. Following the characterisation and genome sequencing of ostreid herpesvirus 1 (OsHV-1), specific diagnostic tools have been developed based on conventional PCR techniques or in situ hybridisation. We have now developed a real-time PCR assay for rapid, sensitive and quantitative detection of OsHV-1, and compared it with a conventional PCR technique described previously. Molecular responses of Ostrea edulis haemocytes to an in vitro infection with Bonamia ostreae ArchiMer

Bonamiosis due to the parasite Bonamia ostreae is a disease affecting the flat oyster Ostrea edulis. B. ostreae is a protozoan, affiliated to the order of haplosporidia and to the cercozoan phylum. This parasite is mainly intracellular, infecting haemocytes, cells notably involved in oyster defence mechanisms. Suppression subtractive hybridisation (SSH) was carried out in order to identify oyster genes differentially expressed during an infection of haemocytes with B. ostreae. The most known and certainly the most important contribution of genetics to the oyster indus try is the development of the triploid cupped oyster. Released for the first time in the 1980s, the triploid oyster has triplets of  chromosomes instead of pairs. Currently, triploid oysters are obtained by crossing diploid and tetraploid oysters. These oysters are characterized by a near lack of milt in summer, which makes them generally popular with consumers, and by a faster growth, allowing a shorter production cycle. Little information is available on viral infections that affect bivalve molluscs. Such a lack of data is due ta a certain inadequacy of the diagnosis methods that are employed when massive mortality events occur. Most laboratories involved in mollusc pathology still analyse samples through light microscopy. World-wide, there is thus currently a lack of information concerning the occurrence of bivalve herpesvituses. This is probably due to the lack of suitable diagnostic tools. Infections à virus de type herpes chez les bivalves : large spectre d'hôte et détection au stade adulte ArchiMer

In 1972, Farley reported the presence of herpes-type viral particles in adult American oysters (Crassostrea virginica) for the first time. Since then, this pathogen has been detected in various marine bivalve species, usually associated with massive mortality events. To date, herpes-type viruses have been detected in the following species: C gigas, Ostrea edulis, O. angasi, Tiostrea chilensis, Ruditapes decussatus, R. philippinarum and most recently, Pecten maximus.

Development of a polymerase chain reaction for the detection of abalone herpesvirus infection based on the DNA polymerase gene ArchiMer

A 5781-base pair (bp) fragment of genomic DNA from the Taiwanese abalone herpesvirus was obtained and showed 99% (5767/5779) homology in the nucleotide sequence and 99% (1923/1926) in the amino acid sequence with the DNA polymerase gene of the abalone herpesvirus strain Victoria/AUS/2007. Homology of the amino acid sequence with the DNA polymerase of ostreid herpesvirus 1 was 30% (563/1856). Le Réseau REPAMO : un outil de surveillance de la santé des mollusques marins élevés et sauvages ArchiMer

The Ifremer Repamo network (REseau de PAthologie des MOllusques) is the national surveillance network in charge of monitoring marine mollusc health on French coastlines. It is commissioned to enforce the official regulations and to provide a public service, under the supervision of the competent authority (DGAL Veterinary Directorate) in the Ministry.of Agriculture and Fisheries. Its goals are to prevent the introduction and the dissemination of infectious agents, in particular pathogens involved in reportable diseases, and to monitor the changes in the pathogens already present on the French coasts.

Mortalities in a number of commercially important mollusc species have been associated with the detection of viruses belonging to several families. The first description of a virus was in adult eastern oyters, Crassostrea virginica, with the detection of virus particles resembling members of the family Herpesviridae. Herpesviruses associated with mortalities among Pacific oyster, Crassostrea gigas, in France-Comparative study ArchiMer

Sporadic mortalities were reported in june and july of 1992 and 1993 among batches of hatchery-reared larval Pacifie oyster, Crassastrea gigas, and at the beginning of July and in August of 1993 among five batches of 3-7 month old young spats, C. gigas. Observations with transmission electron microscope showed the presence of herpes-like virus particles in infected larvae and young spats. The viruses observed in diseased larvae and affected young spat oysters were very similar. Elevated temperature and crowding may increase susceptibility of oysters to these herpes-like virus infections.

In situ localization and tissue distribution of ostreid herpesvirus 1 proteins in infected Pacific oyster, Crassostrea gigas ArchiMer

Immunohistochemistry (IHC) assays were conducted on paraffin sections from experimentally infected spat and unchallenged spat produced in hatchery to determine the tissue distribution of three viral proteins within the Pacific oyster, Crassostrea gigas. Polyclonal antibodies were produced from recombinant proteins corresponding to two putative membrane proteins and one putative apoptosis inhibitor encoded by ORF 25, 72, and 87, respectively.
